We demonstratedthata bound proteaseunlike plasma kallikreinwas involvedinthe activation. Thisactivity was maximally detected at pH value$ intheranges 6-9,and was strongly inhibitedby diisopropylfluoropho$phate and aprotinin, indicatingthattheenzyme responsible forthe activation isa serine protease.insummary, thispaperdescribes forthefirsttimethata proteasecomplexed with a2-macroglobulin exhibits detectableproteolytic activity towardtheproteinsubstrate single-chain tissue-typeplasminogen activator. |
